Talk Abstract: Chromosome instability (CIN), the missegregation of chromosomes during mitosis, is a common and highly malignant feature of cancer cells. The cell cycle protein Emi1 is overexpressed in most human solid tumours and this predicts poor patient outcome. In transgenic mice, we found that Emi1 overexpression promotes the development of a wide range of aneuploid tumours, with mitotic cancer cells showing lagging and missegregating chromosomes. In vitro, Emi1 overexpression promotes CIN following delays in chromosome alignment and anaphase onset. Finally, in human tumors, Emi1 overexpression is a stronger marker for CIN than most well established markers. Thus, we identified overexpression of the cell cycle protein Emi1 as a novel in vivo mechanism of CIN and driver of tumorigenesis. These novel mechanistic insights have important clinical implications.
Speaker's bio: Dr Pascal Duijf received a PhD degree in Human Genetics from the Radboud University Nijmegen in the Netherlands. His postdoctoral training at Memorial Sloan-Kettering Cancer Center in New York focussed on studying the cancer biology and genetics of chromosome instability in cancer development using mouse models. In 2013, he was appointed as a Group Leader in the Cancer Program at UQ Diamantina Institute. His main research interest is in genomic instability in cancer development, which he studies using a broad range of approaches, including in the areas of cancer genomics, mouse modelling, cell biology, molecular biology and pathology.
