Talk Abstract: Bile pigments (BP) such as biliverdin, bilirubin, protoporphyrin and their derivatives have recently been shown to inhibit the mutagenic effects of agents such as Aflatoxin B1 (AFB1), 2-aminofluorene (2AF), benzo[]pyrene (B[]P) and 2-amino-1-methyl-6-phenylimido[4,5-b]pyridine (PhiP). The mechanism of this inhibition is, however not understood. Our project aims to explore physical, chemical or enzymatic mechanisms that might explain the observed inhibition of mutagenesis. Possible --interactions between the polyaromatic mutagens and the BPs were examined using spectroscopic methods but no significant interactions were detected. The possibility of the BPs acting as chemical scavengers was investigated by exhaustive studies of the reactivity of a model aromatic epoxide, styrene oxide, with various BPs. While some covalent adducts have been identified, it is unlikely that this is the sole mechanism attributable to the inhibition these compounds exhibit against environmental mutagens. Finally, we will present evidence that certain BPs are inhibitors of the P450 enzymes responsible for the activation of the polyaromatic compounds to their highly reactive mutagenic metabolites.
Speaker's bio: Associate Professor Joanne Blanchfield did her PhD at UQ with Prof. Bill Kitching then completed postdoctoral positions at Johns Hopkins University with Prof. Craig Townsend and at Australian National University with Prof. Lew Mander. Upon returning to UQ she worked in the School of Pharmacy with Prof. Istvan Toth before being appointed as a lecturer in SCMB in 2003. Her main areas of research include the isolation and bioavailability of natural products and the design and synthesis of components of synthetic vaccines.
