Talk Abstract: : Genome-wide association studies (GWAS) have identified a large number of genetic variants associated with human complex traits. However, genes or functional DNA elements through which these variants exert their effects on the traits are often unknown. I will present a method (called SMR) that integrates summary-level data from GWAS and eQTL studies to identify genes whose expression levels are associated with a trait. An application of the method to large data sets for five complex traits identifies 126 genes, of which 77 genes (~61%) are not the nearest genes of the GWAS top SNPs. I will then show how this method can be extended to include other types of omics data to pinpoint the functional elements where the causal variants are possibly located. The method and results are helpful for the design of follow-up functional studies to understand the genetic basis of complex disease.
Speaker's bio: Jian Yang is a Professor at Institute for Molecular Bioscience, The University of Queensland. He received his PhD in 2008 from Zhejiang University, China. His research interests are in developing novel methods and tools to better understand the genetic architecture of complex traits and disease using high-throughput genomic data. He won the Centenary Institute Lawrence Creative Prize in 2012, and was one of two recipients of the Sylvia and Charles Viertel Charitable Foundation's Senior Medical Research Fellowship in 2014. He was awarded the Australian Academy of Science Ruth Stephens Gani Medal for distinguished research in human genetics in 2015.
